益气活血利水法治疗心力衰竭水潴留机制的实验研究

【摘 要】目的:探讨益气活血利水法治疗心衰水潴留的机制。方法:选择健康Wistar大鼠,应用腹腔注射阿霉素,造成心衰大鼠模型,然后分模型组、西药对照组,中药对照组、中药高剂量、中剂量、低剂量6组,给药8周后观察各组大鼠服药后血管加压素(AVP)浓度;血管加压素V2受体(V2R)、水通道蛋白2(AQP-2)蛋白表达及mRNA表达情况。结果:益气活血利水法可降低心衰大鼠血浆中AVP浓度,减弱肾组织V2R、AQP-2蛋白表达,降低V2R、AQP2、mRNA相对表达量。结论:益气活血利水法通过作用于精氨酸血管加压素-V2受体-水通道蛋白2系统的各个环节而起到改善水液潴留的作用。

【Abstract】Objective:To investigate the Yiqi Huoxue Lishui therapy for the water retention mechanismof heart failure. Methods: Healthy Wistar rats, with intraperitoneal injectionof doxorubicin, resulting in a rat model of heart failure, and then were divideinto the model group, western medicine group,TCM control group, high dose,medium-dose,low dose six groups administered 8 weeks after the rats aftertaking vasopressin (AVP) concentration; vasopressin V2 receptor (V2R),aquaporin-2 (AQP-2) protein and mRNA expression. Results Yiqihuoxue therapy can reduceheart failure in rat plasma AVP concentration, decreased kidney tissue V2R, AQP-2protein, lower V2R, AQP2,mRNA relative expression.Conclusion: Yiqihuoxue therapyby acting on arginine vasopressin receptor -V2 - all aspects of aquaporin 2system and play a role in improving water retention.
