兔骨髓基质细胞源性神经干细胞诱导分化及 移植修复脊髓损伤实验研究

【摘  要】目的：探究骨髓基质源性神经干细胞对于修复脊髓损伤的可行性和可靠性。方法：采用新西兰白兔作为研究对象，提取骨髓基质细胞加以培养分化，手术方式将骨髓基质细胞源性神经干细胞悬浊液移植到脊髓损伤部位，观察损伤区域再生情况。结果：早期培养细胞在无诱导分化时能表达巢蛋白，表明其有干细胞活性；干细胞移植后脊髓损伤部位增殖细胞排列无层次，且BrdU染色呈阳性。结论：兔骨髓基质细胞在合适的体外培养条件下能分化成干细胞，具有增殖分化能力，可以应用于修复脊髓损伤等神经学科领域。

【Abstract】Objective: To explore the feasibility andreliability of bone marrow stromal derived neural stem cells in repairingspinal cord injury. Methods:New Zealandrabbits were used as the research objects to extract bone marrow stromal cells,which were given culture differentiation, operation was made to transplant thesuspension of the bone marrow stromal derived neural stem cells to the site ofspinal cord injury and the regeneration in the damaged area was observed.Results: The early cultured cells in no induced differentiation could expressnestin, which proved the activity of stem cells; there was no hierarchicalarrangement of cell proliferation in spinal cord injury parts after stem celltransplantation and BrdU staining was positive.Conclusion:Rabbit bonemarrow stromal cells could differentiate into stem cells in vitro; it had theability of proliferation and differentiation, which could be used in the fieldof spinal cord injury.
