【摘 要】目的:探讨阿魏酸钠(sodiumferulate, SF)对溃疡性结肠炎治疗的可能相关机制。方法:小鼠随机分4成组空白对照(NC)组、模型(MD)组、低剂量(SFLD)、高剂量(SFHD)阿魏酸钠治疗组,NC组采用正常饮用水饲养,采用5%葡聚糖硫酸钠(Dextransulfate Sodium, DSS)制作UC小鼠模型,造模7 d成模后分为MD组、SFLD组和SFHD组,予药物干预治疗后续7 d。记录分析疾病活动指数(DAI)及脾脏指数(SpleenIndex SI),取结肠组织行HE染色;Westernblot检测结肠NF-KB表达水平;ELISA法测小鼠肠粘膜NF-kB活性。结果:SFLD组、SFHD组小鼠第8~14 d DAI明显低于MD组(P<0.05);SFLD组、SFHD组小鼠第14 d SI明显低于MD组(P<0.05);MD组SI明显高于NC组(P<0.05);SFLD组、SFHD组结肠NF-kB表达和活性明显低于MD组;MD组结肠NF-kB表达和活性明显高于NC组(P<0.05);MD组NF-kB表达和活性明显高于NC组(P<0.05)。结论:阿魏酸钠可以通过降低结肠NF-kB表达及活性;抑制脾脏代偿性增大,从而减轻肠道的炎症反应及抑制脾脏的免疫反应,改善UC全身炎症状态及免疫应答,对UC发挥治疗作用。

【Abstract】Objective:To investigate the mechanism of sodium in thetreatment of ulcerative colitis. [Method] Mice were randomly divided into 4groups: blank control (NC) , model group (MD group), low dose SF group(SFLD), high dose SF group(SFHD). NC group withnormal drinking water feeding, After 7 days of using 5% dextran sulfate sodiumto make UC mouse model, the models with 7 days of drug intervention weredivided into MD group, SFLD group and SFHD group. Record and analyze thedisease activity index (DAI) and spleen index (Spleen Index SI), colon tissuewere stained with HE ; NF-KB expression of colon were detected by western blot;NF-kB activity of intestinal mucosal were measured by ELISA. Result:Among SFLD group and SFHDgroup of mice on days 8-14, DAI was significantly lower than that of MD group (P<0.05); Among SFLD group and SFHDgroup of mice on 14 days, SI was significantly lower than that of MD group(P<0.05); the SI of MD group wassignificantly higher than that of group NC (P<0.05);Among SFLD group and SFHD group, the expression and activity of NF-kB in colonwas significantly lower than that of MD group; the active of NF-kB expressionin MD group was significantly higher than that in NC group (P<0.05); the activity of NF-kBexpression in MD group was higher than that in NC group (P<0.05). Conclusion:Sodium ferulate can reduce the expressionand activity of NF-kB of colon, inhibit the spleen’s compensatory increase,relieve the inflammatory reaction of intestine,alleviate immune reaction ofspleen,improve systemic inflammatory state and immune response of UC and play arole in the treatment of UC.
